Thanks to GHammer first for making me think about the subject and for inspiring me to do the research into everything - I wrote whole theory down on his post about the healing process, but I think a lot of people are unlikely to read it and get involved unless I made a whole new post.
Im sorry its so long, but If I go into quite a bit of detail people who have good Penis Enlargement knowlege and can then help thorise and make Penis Enlargement a more scientific experience and get some scientific proof that it works/improve gaining rates..
General Anatomy and Macrostructure
The penis is made of smooth muscle among other things.. It's a little bit like skeletal muscle (biceps etc.) - because it contains the same molecules - but it is arranged much differently, is not excited by neuronal stimulation and contracts slightly differently.
Briefly: If weightlifiting - the increase in muscle size is due to lifting heavier weights which causes the following:
The number of sarcomeres increases and the distance between M-line and Z-disk decreases.
So you pack more muscle into the space you orignially have by making things more compact and shoving more in.
(Think a line of beer cans - if you weightlift - you crush all these cans into shorter ones and add more shorter ones on the end to keep the line of cans the same length - but there would be many more cans in the line after crushing them)

In relation to the penis:
Ive looked at a few articles on google about smooth muscle growth - and what a surprise.. not much is known.
But there is some interesting information about how they have grown vascular smooth muscle (whats in the cock) which was put under constant mecanical stress for 48 hours.
And in 48hours there was an overall 40% increase in the number of cells.
This was done on glass plates though and is only one of the types of cell present in the penis.
There was probably also a selection of growth factors (hormones) specifically for the promotion of the growth of this tissue.
As for the other tissues in the penis:
If you (for some reason) really really badly wanted to do the splits it would be wise to take it slow to avoid injury.
If you were really commited 20minutes a day of stretches and eventually you would get there. (I understand that the anaolgy uses tendons instead of ligaments but the difference is not massive, apart from in the above analogy there would also be a level of skeletal muscle stretch which would make it easier)
You literally cant do it over night without breaking something.
So taking the above information - for growth of smooth muscle I think it is not -as- required to take a break for more than 24 hours in comparison to the other cell types down there.
(Note: cell death was not measured in the study)
But for all the other tissues in the penis - I would say may need a bit more time to adjust to the stresses - to avoid damage.
(Think skiiers or super-obese (is that what its termed?) people that over do it and damage their cruciate ligament.
Most importantly with gains you must Listen to your penis:
So, If it hurts then stop doing it (obviously!) proliferation rates can be quite remarkable, the body is well designed to adjust to stess. If the stretches are heavy you'll need more time for the cells to proliferate and heal compared to light stretches.
So.. How does the above effect Penis Enlargement?
I personally think you could find a level at which you stretch everything and only need the hours from sleep to recover which would give you the best gains. - No specific recovery time and working to a maximum injury free level. (I feel the same for bodybuilding - and am experimenting at this moment with both Penis Enlargement and bodybuilding)
I would suggest the older guys here took more breaks though because they are at more risk of ligament damage... and Its supposed to be very painful.
This leads me to believe that you COULD do stretching once or even twice a day (say first thing in the morning and last thing at night) but it would down to how much stress you deliver to your penis.
You would need to find "the line" between overdoing things and being within your confort zone - then train on that line but not going over it.
If youre at "the line" or below, 8 hours SLEEP or 14+ hours AWAKE should be long enough to recover .
The level at which you exceed "the line" alters how much longer you need to recover.
For example:
You could do either 2 sets of light 20 minute jogs (one in the morning one in the evening separated by 14ish hours) or a long 40 minute jog.
The second COULD and think does take longer to recover from due to drastically increased macro and microdamage (lactic acid build up, low glycogen stores and muscle damage) compared to the first.
But WHICH one provides the best gains?
Per singular session I would guess the 40minute jog but this would require a longer recovery time.
Overall, I would vote the 2x20 mins session jog because you do not need to spend time in recovery!
You may find that doing 2x21 minute jogs leads to slight soreness which requires more recovery time - potentially reducing gains.
(Okay this is still jogging and not Penis Enlargement - but the principal is the same)
Another Idea - which follows on. - Keep with it! Almost done.
This is utilising the idea(I think it is a fact..) that recovery rates are higher during sleep than during the day...
After a morning session because you are unlikely to ever fully recover before the evening session you could force growth to be faster.
Think bone stretching procedures:
You make a break in the bone
Just as the bone starts to bind, you could make that length just a little bit bigger.
And everytime, just before it binds you make it a little bit further away.
(repeated partial recovery)
Opposed to making a break and just letting it heal.
(full recovery)
And rebreaking it.
Clearly the 1st situation is much faster - but still relatively slow.
So how does this affect Penis Enlargement?
You could do twice as much work in an 8 day period, and have added benefits of no recovery time.
2 x 20minute session - 7days a week
instead of
4 x 40minute session - 3/4 days a week
[The benefits which I mention are the idea of constantly increasing the gap between two cells and not allowing the gap to be fully closed - full recovery]
I think that bone stretching theory is similar to any tissue stretching.
How does this relate to Penis Enlargement gains?
Girth excercises/compression (e.g. jelqing) would promote smooth muscle growth a lot more than stretching because smooth muscle is what fills with blood during an is stretched during a jelq.
Smooth muscle cells grow quite rapidly (according to our rat tissue) and is what causes improved Erection Quality (EQ) to be one of the first things we noitce when Penis Enlargementing.
Length and girth however requires increased tunica and ligament tissue etc. which takes a longer amount of time - (Think bone stretching)
Length improves faster if you stretch the ligaments as well as the tunica
And girth improves from just tunica stretching.
However - without smooth muscle to fill the tunica errection quality is likely to reduce unless PC muscle is good.
