I'd definately have to pick some form of girth exercise, as that's what I'm having the most trouble attaining...as for which one, that's a difficult decision. Probably cable clamp squeezes.

Wet, dry, up, down, and sideways, it's got something for every aspect of the cock. I learned this exercise as my first and it still seems to me the absolutely essential NPenis Enlargement building block.

I'm still a newbie, being on phase 1. It's hard for me to say which exercise produces the most gains, but I know I get the biggest pump from jelqing.
 
If we were talking about pre-Penis Enlargement I would have to vote for Jelqing.

However, if we were talking about current status I would vote for manual stretches.
