I was looking up if there was a way to induce a 2nd puberty localized to the penis. During puberty testosterone production increases and the testosterone links with Androgen receptors which signal the Corpora Cavernosa, Tunica Albunginea, ligaments, and surrounding penile tissue to grow. By the end of puberty Androgen receptors stay open to testosterone but they stop sending growth signals. The ultimate advancement in PE would be to find a way to reopen those growth signals, but localized to only the penis, without also growing cancerous cells. Then being able to shut it off.

Ah, the Golden Egg question that has been asked since 1970s. You've nailed it on the head and it was and still is under research for the past 5 decades. HGH was used locally to induce growth, and synthetic androgens (AAS) in the modern days also used on desperate patients that would go to great length of any kind for growth beside subjecting under the knives and implants.
Androgen receptors constantly remained opened, but at a lower level. Your penis is the same way, as long as you induce sufficient amount of stress loads accordingly to excite the androgen receptors to reactivate during times of needs. I always emphasize safety because that's how the body works. When the body knows there's not much stresses elsewhere, plenty of bioavailable resources at the ready, and small damages occurs around the penile area, the body diverts its resources for growth little by little, and for a few, it triggers secondary and tertiary puberty growth with phenomemal gains. Even when we're in our old ages, we still see gains because the androgen receptors are not fully closed or deactivated. We just need to trick the body to reactivate at a higher level.
As you said, using of the anabolic-androgenic steroids (AAS) in a localized area can bring out irregular growths, such as cancer. This does not mean you cannot use a very low dosage to "boost" receptors' awareness and reactivation, stimulating a puberty-like phase in the cells. There has been multiple test cases in using cocktails of Insulin-like growth factor-1 (IGF-1) with AAS in place of HGH. Unfortunately, part of the world do not allow such use of AAS, IGF-1, and HGH without proper medical prescriptions and medical monitoring. There are plenty of cases of regions in the world that will allow uncontrolled use of the cocktail, and of course, people do order the substances in using them as illegal and illicit products.
The final thought is, do the cocktails or applications of the synthetic compounds work? The answer is, 50/50. It did work for a few, while other reported it them horny but no growth. A couple case did report that the prostrates and gonads gotten larger as a result. The gonads were not an issue, but the enlarged prostates were of concerns.
For those who actually gained, they did gain 1in to 1.5in without having to do any PE. For those who use in conjunction, they did gain up to 2.5in. The timelines were skewed due to the reporting periods they came back with, anywhere between 1.2 to 2.5 years.
